Vinkristine maruz kalmış ratlarda tribulus terrestrisin sperma kalitesi ve testis toksisitesi üzerine etkileri

Abstract

Bu çalışma, kemoterapötik ajan vinkristin maruziyetinin erkek Wistar-Albino ratlarda üreme sistemi üzerindeki olumsuz etkilerini araştırmakta ve bu etkilerin önlenmesinde bitkisel bir ajan olan Tribulus terrestris 'in potansiyel koruyucu etkilerini incelemektedir. Çalışmada, spermatolojik ve histolojik parametreler, DNA hasarı, oksidatif stres göstergeleri ve serum testosteron seviyeleri değerlendirildi. Araştırma, 45 gün boyunca kontrol, yalnızca Tribulus terrestris uygulanan, Tribulus terrestris ile vinkristin birlikte uygulanan ve yalnızca vinkristin verilen rat gruplarında yürütüldü. Vinkristin uygulaması, seminifer tübüllerin çapında ve epitel yüksekliğinde azalma, spermatozoon motilitesi ve sayısında düşüş, DNA hasarı ve histolojik bozukluklarla sonuçlandı. Tribulus terrestris uygulaması ratlada antioksidan ve hücresel koruyucu etkileriyle vinkristin kaynaklı bu zararları anlamlı ölçüde azalttı. Tribulus terrestris'in seminifer tübül sağlığını koruduğu, spermatoza kalitesini artırdığı ve DNA hasarını önlediği gösterildi. Tribulus terrestris'in etkileri, muhtemelen androjen salınımını artırması ve hücresel antioksidan savunmayı güçlendirmesiyle ilişkilendirili olduğu kanaatine varılmıştır. This study investigates the adverse effects of the chemotherapeutic agent vincristine on the reproductive system of male Wistar-Albino rats and examines the potential protective effects of the herbal agent Tribulus terrestris in mitigating these effects. Parameters such as spermatological and histological changes, DNA damage, oxidative stress markers, and serum testosterone levels were evaluated. The experiment was conducted over 45 days with rat groups divided into control, Tribulus terrestris only, combined Tribulus terrestris and vincristine, and vincristine-only groups. Vincristine exposure resulted in reduced seminiferous tubule diameter and epithelial height, decreased sperm motility and count, increased DNA damage, and histological disruptions. Administration of Tribulus terrestris significantly alleviated these damages in rats through its antioxidant and cellular protective effects. It was demonstrated that Tribulus terrestris preserved seminiferous tubule health, improved sperm quality, and prevented DNA damage. The effects of Tribulus terrestris were attributed to its potential to enhance androgen release and strengthen cellular antioxidant defenses. This study highlights the protective potential of Tribulus terrestris against the toxic effects of vincristine on the reproductive system and suggests its use as a supportive agent during chemotherapy.
